A Temperature-Tolerant α-Amylase and Its Degradation Research on Oilfield Starch-Based Polymers

摘要:The enzymes to alleviate polymer plugging is a time-honored and effective strategy in drilling, completion, hydraulic fracturing and enhanced recovery (EOR) operations. In this study, a temperature-tolerant α-amylase was characterized based on comprehensive characterizations. The HPLC and FTIR tests showed that α-amylase be consists of 17 amino acids, and the molecular chain contains a variety of functional groups such as -COOH and peptide bonding (-CO-NH-). The TGA and PSD experiments revealed that α-amylase has multiple weight loss temperatures and particle size intervals, with the most pronounced weight loss at 92 °C, and the charge was slightly biased towards the negative potential in the physiological environment of pH = 7. Rheological testing of the polymers showed that α-amylase was able to significantly degrade three common oilfield starch-based polymers, including carboxymethyl starch (CMS), hydroxypropyl starch (HPS), and heat-resistant modified starch (FLO), at temperatures above 90 °C. In addition, α-amylase can effectively eliminate polymer filter cake in non-solid and temporary plugging water-based drilling fluid systems, reducing viscosity by more than 60% and increasing filtration loss by 2 ~ 9 times. This study provides valuable insights and guidance for the use of bio-enzymes to mitigate reservoir damage and the advancement of microbial enzymes for enhanced recovery. ?
